GİRİŞ ve AMAÇ: Bu çalışmada Porphyromonas gingivalis ATCC 33277, Prevotella intermedia ATCC 25611 ve Aggregatibacter actinomycetemcomitans ATCC 43718 periodontal patojenlerine karşı gıdalarda bulunan fitokimyasallardan kuersetin, kurkumin, apigenin, pirogallol, gallik asit, luteolin ve hidroksitirozolün in vitro antimikrobiyal potansiyellerinin değerlendirilmesi amaçlanmıştır. YÖNTEM ve GEREÇLER: Çalışmada agar kuyucuk difüzyon ve mikrodilüsyon yöntemleri kullanılarak fitokimyasalların antimikrobiyal potansiyelleri araştırılmıştır. Agar kuyucuk yönteminde fitokimyasalların antimikrobiyal aktiviteleri 3 mg/ml, mikrodilüsyon yönteminde ise 3, 1.5, 0.75, 0.375, 0.187, 0.093, 0.046, 0.023 ve 0.011 mg/ml konsantrasyonlarda incelenmiştir. Pozitif ve negatif kontrol olarak amoksisilin (20 µg/disk ve 0.04-0.0015 mg/ml) ve DMSO (≤%10) test edilmiştir. BULGULAR: Çalışma sonuçlarına göre fitokimyasalların çoğunun patojenlere karşı antimikrobiyal aktivite gösterdiği ve patojenler üzerinde en etkili fitokimyasalın kuersetin olduğu belirlenmiştir. Kuersetin, kurkumin, luteolin, apigenin ve gallik asit tüm patojenlere karşı en iyi antimikrobiyal potansiyeli olan fitokimyasallar olarak belirlenmiştir. Çalışmamızda %4 (v/v) ve daha az orandaki DMSO’nun patojenler üzerinde etkisiz olduğu tespit edilmiştir. TARTIŞMA ve SONUÇ: Periodontal hastalıkların tedavisinde antibiyotik kullanımına ilave olarak, özellikle kurkumin, kuersetin, luteolin, gallik asit ve apigenin içeren gıdaların tüketimi uygun ve yararlı olabilir. Bu yüzden, ileride, güçlü antimikrobiyal ve antioksidan potansiyeli olan fitokimyasalların periodontal patojenlerin inhibisyon mekanizmalarına yönelik etkilerinin daha detaylı araştırılması ve bu fitokimyasalların etkin kullanımlarını belirlemek için ileri boyutta in vivo çalışmalar yapılması önerilmektedir.

INTRODUCTION: In this study, it is aimed to investigate antimicrobial potentials of the dietary phytochemicals including quercetin, curcumin, apigenin, pyrogallol, gallic acid, luteolin and hydroxytyrosol against the periodontal pathogen bacteria including Porphyromonas gingivalis ATCC 33277, Prevotella intermedia ATCC 25611 and Aggregatibacter actinomycetemcomitans ATCC 43718. METHODS: The antimicrobial potential of the dietary phytochemicals was investigated by using agar well diffusion and microdilution methods. The antimicrobial activities of the phytochemicals in concentration of 3 mg/ml and in concentrations of 3, 1.5, 0.75, 0.375, 0.187, 0.093, 0.046, 0.023 and 0.011 mg/ml were studied by agar well diffusion and microdilution methods, respectively. Amoxicillin (20 µg/disc, 0.04-0.0015 mg/ml) and DMSO (≤10%) were tested as positive and negative controls. RESULTS: According to the results of this study, the most of the phytochemicals had antimicrobial activity aganist all of the bacteria. The most effective phytochemical against the bacteria was quercetin. The phytochemicals with the strongest antimicrobial potential were quercetin, curcumin, luteolin, apigenin and gallic acid. DMSO (≤4%, v/v) showed no antimicrobial effect on the pathogens. DISCUSSION AND CONCLUSION: In addition to the usage of antibiotic, the consumption of the phytochemicals especially including curcumin, quercetin, luteolin, gallic acid and apigenin could be a suitable and helpful in the treatment of periodontal diseases. Therefore, it is suggested that more research about the inhibition mechanisms of the periodontal pathogens by powerful phytochemicals should be performed and, further extent in vivo studies should be carried out to detect effective usage of these phytochemicals.
